The LIM kinases are a group of 40 proteins which play an important role in cell signalling and growth control. Antibody studies on LIMK-1 and 2 proteins shown they control morphogenesis and cell motility by regulating cofilin activity and the stabilization of F-Actin structures. Recently, Scott et al used LIMK1/2 antibodies to show the protein could play an important part in tumour cell metastasis.

Cancer antibodies are widely used in research into metastasis, as the majority of cancer cells are able to metastasize.

Tumour cell motility requires two types of protein: path generators, which remodel the extracellular matrix (ECM) and lead metastasis, and path followers. Scott et al compared the activity of LIMK1/2 in MDA-MB-231 breast cancer cells and non-tumour stromal cells, to investigate its role in path generation and collective cell invasion. The proteins were blocked using a potent small-molecule inhibitor or Si (short-interfering) RNA-knockdown. Cell migration and invasion were observed using Matrigel as the medium.

It was seen that LIMK inhibition led to reduced cofilin phosphorylation, F-actin levels, transcriptional activity of serum response factor and collagen contraction. Although LIMK knockdown left cell motility unimpaired, the ability to lead strands of collectively invading cells was lost. The ECM degradation, MM9 activation and invadopodia formation necessary for cell invasion could not take place. LIMK inhibition of cancer-associated fibroblasts also led to loss of lead ability in squamous carcinoma cells.

This suggested LIMK is necessary for matrix remodelling and ECM degradation by leader cells, and therefore the path generation and collective invasion of cancer cells in metastasis.
